The play is about two people who wake up next to each other the morning after attending a wedding.
The Way We Get By will be the New York stage debut for the Regina native, who appeared in theatre productions as a child and while attending Dr. Martin LeBoldus High School.
Previews begin May 12, 2015 and the play is scheduled to run to June 28.
Maslany’s work on Orphan Black earned her a Canadian Screen Award and two consecutive Critics’ Choice Television Award as well as nominations at the Golden Globes, ACTRA Awards, People’s Choice Awards and Dorian Awards.
She previously won Gemini Awards for guest roles on Flashpoint and Bloodletting and Miraculous Cures.
